Primovir Nirmatrelvir is an important protease inhibitor of SARS-CoV-2. It works by preventing the growth of the virus that causes COVID-19. Ritonavir increases (“boosts”) levels of nirmatrelvir. This helps nirmatrelvir work better. For more information about nirmatrelvir and ritonavir, see the FDA Emergency Information Sheet for Patients, Parents, and Carers and Health Canada Drug Patient Information Sheet.

Information about how safe and effective the combination of nirmatrelvir and ritonavir is to treat COVID-19 is limited. Study results suggest that Primovir nirmatrelvir and ritonavir can help people who have recently tested positive for the coronavirus stay away from the hospital. This product is not for use by people who are hospitalized with COVID-19.

The information in this document reflects new, evolving data and is subject to re-evaluation. Users should be aware of these considerations in their reviews of nirmatrelvir and ritonavir, and it is always the responsibility of the treating physician to exercise independent judgment in making treatment decisions.

Ritonavir is used with nirmatrelvir because it acts as a pharmacokinetic enhancer, resulting in higher systemic concentrations and a longer half-life of nirmatrelvir, thereby maintaining a twice-daily dosing regimen.
